After extraction, lactose and sucrose in the sample are separated using a high-performance liquid chromatography column and detected using a differential refractive detector or evaporative light scattering detector. The external standard method is used for quantification. Unless otherwise specified, all reagents and materials used in this method are analytical pure, and the water is Grade 1 water as specified in GB/T 6682. 4.1 Acetonitrile. 4.2 Acetonitrile: Chromatically pure. 4.3 Standard solution 4.3.1 Lactose standard stock solution (20 mg/mL): Weigh 2 g of lactose standard sample (accurate to 0.1 mg) that has been dried in a 94 ℃ soil 2 ℃ oven for 2 hours, dissolve it in water, and dilute with water to a 100 mL volumetric flask. Place in a 4 ℃ refrigerator. 4.3.2 Lactose standard working solution: Take 0 mL, 1 mL, 2 mL, 3 mL, 4 mL, and 5 mL of lactose standard stock solution (4.3.1) into a 10 mL volumetric flask, and bring to volume with ethylene wax (4.1) to the mark. Prepare a series of lactose standard working solutions with concentrations of 0 mg/mL, 2 mg/mL, 4 mg/mL, 6 mg/mL, 8 mg/mL, and 10 mg/mL. 4.3.3 Sucrose standard solution (10 mg/mL): Weigh 1gK (accurate to 0.1 mg) of sucrose standard sample dried in a 105 ℃ soil 2 ℃ oven for 2 hours, dissolve it in water, and dilute with water to a 100mL volumetric flask. Place in a 4 ℃ refrigerator. 4.3.4: Sucrose standard working solution: Take 0 mL, 1 mL, 2 mL, 3 mL, 4 mL, and 5 mL of sucrose standard solution (4.3.3) into a 10 mL volumetric flask, and bring to volume with acetonitrile (4.1) to the mark.
