Amino acid detection method:1. Weigh the sample
After crushing, accurately weigh the sample with an electronic balance to the nearest 0.0001g (within the protein content range of 10mg~20mg), and place the weighed sample in a hydrolysis tube
2. Hydrolysis
Add 15mL of 6mol/L hydrochloric acid to the hydrolysis tube, 3-4 drops of newly distilled phenol, and then place the hydrolysis tube in ice water, freeze for 3-5 minutes, and connect it to the vacuum pump's suction pipe. After vacuuming, flush in high-purity nitrogen gas; Vacuum and fill with nitrogen again, repeat three times, and seal in a nitrogen filled state. Place the sealed hydrolysis tube in a constant temperature drying oven at 110 ℃ and 1 ℃. After hydrolysis for 22 hours, remove and cool it
Open the hydrolysis tube, filter the hydrolysate, rinse the tube multiple times with deionized water, transfer all the hydrolysate to a 50mL volumetric flask, dilute with deionized water, draw 1mL of the filtrate into a 5mL volumetric flask, dry with a vacuum dryer at 40 ℃~50 ℃, dissolve the residue in 1-2 mL of water, and then dry again. Repeat twice before evaporating to dryness. Dissolve in 1mL of pH2.2 buffer solution for instrument measurement
